Control of tobacco mosaic disease by combined application of the resistance inducer fTDP and ningnanmycin

    【目的】fTDP蛋白(fusion TEER-decreasing protein, fTDP)可诱导烟草对烟草花叶病毒(Tobacco mosaic virus, TMV)产生抗性,为提高fTDP蛋白的田间防效及稳定性,本研究拟筛选复配剂确定最佳配比,并探索其诱导机制。【方法】以三生烟和普通烟K326为材料分析不同药剂处理后烟草枯斑数,染病症状以及TMV外壳蛋白基因(TMV-CP)表达水平等,筛选出抗病毒效果最强的药剂;利用共毒系数法确定所选药剂宁南霉素与fTDP蛋白最佳配比,并从植株生长状态、防御酶和防御基因等指标,评价fTDP蛋白/宁南霉素复配剂的抗病效果和抗性途径;以云烟87为材料测定复配剂田间应用效果。【结果】宁南霉素与fTDP蛋白能有效延缓烟草的花叶症状并显著降低病毒含量。当fTDP蛋白与宁南霉素质量比为3∶7时,复配剂对TMV钝化作用相对单剂增效显著,共毒系数达152;在fTDP蛋白与宁南霉素质量比为1∶9时,复配剂可保护烟草K326不被TMV侵染,相对单剂增效显著;复配剂也可提高烟草K326水杨酸含量,防御酶活性以及多种抗病相关蛋白基因的表达水平;田间试验结果显示,该配比下的复配剂比单剂具有更高更稳定的药效,防效可达77.8%。【结论】fTDP蛋白/宁南霉素复配剂可通过激活水杨酸信号通路介导的系统诱导抗性提高烟草对TMV的抗性,fTDP蛋白与宁南霉素质量比为1∶9时,复配剂在室内外抗病毒活性和降低成本方面均优于单一药剂。

    Abstract:

    【Aim】The fusion TEER-decreasing protein (fTDP) induces tobacco resistance to tobacco mosaic virus (TMV). To enhance the field efficacy and stability of fTDP, we optimized the combination ratios of agents and explored the induction mechanism. 【Method】Using Nicotiana tabacum var. Samsun NN and N. tabacum K326 as materials, we analyzed the number of necrotic spots, disease symptoms, and expression levels of the TMV coat protein gene (TMV-CP) in tobacco plants treated with different agents to identify the most effective antiviral agent. The optimal ratio of the selected agent, ningnanmycin, to fTDP was determined via the co-toxicity coefficient method. The disease resistance efficacy and resistance pathways of the fTDP/ningnanmycin combined agent were evaluated based on indicators such as plant growth status, defense enzymes, and defense genes. The field application efficacy of the combined agent was measured with Yunyan 87 as the material. 【Result】Ningnanmycin and fTDP delayed the appearance of mosaic symptoms of tobacco and significantly reduced the viral load. When the mass ratio of fTDP to ningnanmycin was 3 : 7, the combined agent exhibited a significant synergistic effect of TMV inactivation, with a co-toxicity coefficient of 152. When the mass ratio of fTDP to ningnanmycin was 1 : 9, the combined agent protected tobacco K326 from TMV infection, with a significant synergistic effect. In addition, the combined agent increased salicylic acid content, defense enzyme activity, and expression levels of multiple disease resistance-related protein genes in tobacco K326. Field trial results demonstrated that the combined agent at this ratio exhibited higher and more stable efficacy than the single agents, achieving the control efficacy of 77.8%. 【Conclusion】The fTDP/ningnanmycin combined agent enhances tobacco resistance to TMV by activating the salicylic acid signaling pathway. When the mass ratio of fTDP to ningnanmycin was 1 : 9, the combined agent outperformed single agents in both antiviral activity and cost reduction in both laboratory and field settings.
